Not sure about optimal, but on a clean build the defaults are:

Fonts are "Sans" and "Sans bold"
201 dpi
Smoothing = Grayscale
Hinting = Slight

I suspect these defaults are also the optimum.

No.  Subpixel smoothing will only reduce quality when used on an XO
laptop.  Subpixel smoothing relies on the particular geometry of a
standard laptop display, and does not work on OLPC's Pixel Qi displays.
